i work on semis for a living and those trannys are massave compaired to the ones in our pickups. another thing you will need is constant onboard air. they shift via air pressure and if ur air pressure is too low or drops too low it wont go into gear

There are many things to consider. I saw an old school with a 10spd and there was boku custom parts on board.

That would be and awesome project. Dont know what it would take to make it happen though but would be fun. I seen a vid on youtube a k30 chevy club cab like a late 80's model with a cummins swap and a 10spd behind it so It can be done but what it would take IDk. Good luck and would be a very interesting thread to follow on here if you do go for it.
